The angiotensin I-converting enzyme (ACE) inhibitory activities of protein hydrolysates prepared from heads and viscera of sardinelle (Sardinella aurita) by treatment with various proteases were investigated. Protein hydrolysates were obtained by treatment with Alcalase®, chymotrypsin, crude enzyme preparations from Bacillus licheniformis NH1 and Aspergillus clavatus ES1, and crude enzyme extract from sardine (Sardina pilchardus) viscera. All hydrolysates exhibited inhibitory activity towards ACE. The alkaline protease extract from the viscera of sardine produced hydrolysate with the highest ACE inhibitory activity (63.2 ± 1.5% at 2 mg/ml). Further, the degrees of hydrolysis and the inhibitory activities of ACE increased with increasing proteolysis time. The protein hydrolysate generated with alkaline proteases from the viscera of sardine was then fractionated by size exclusion chromatography on a Sephadex G-25 into eight major fractions (P1–P8). Biological functions of all fractions were assayed, and P4 was found to display a high ACE inhibitory activity. The IC50 values for ACE inhibitory activities of sardinelle by-products protein hydrolysates and fraction P4 were 1.2 ± 0.09 and 0.81 ± 0.013 mg/ml, respectively. Further, P4 showed resistance to in vitro digestion by gastrointestinal proteases. The amino acid analysis by GC/MS showed that P4 was rich in phenylalanine, arginine, glycine, leucine, methionine, histidine and tyrosine. The added-value of sardinelle by-products may be improved by enzymatic treatment with visceral serine proteases from sardine. 相似文献

In this study, fruit and vegetable by-product and waste sources in Ireland were tested for their antioxidant activity and polyphenol content. The highest levels of antioxidants measured by both ferric reducing antioxidant power (FRAP) and diphenyl-picrylhydrazyl (DPPH) assays, were detected in whole kiwifruit. Of the vegetable by-products, broccoli stems showed the best antioxidant potential. The level of polyphenols as assessed by the Folin–Ciocalteu Reagent (FCR) was significantly correlated with the level of polyphenols by HPLC–DAD (r = 0.93). The level of polyphenols assessed by HPLC–DAD was not significantly correlated with the antioxidant assays. Apple pomace and vegetable by-products were found to be good sources of both polyphenols and antioxidants and due to their abundance may be exploitable resources to use as food ingredients. 相似文献

Muscle proteins were recovered from rainbow trout processing by-products (fish meat leftover on bones, head, skin, and etc.) by isoelectric solubilization/precipitation. Muscle proteins precipitated at pH 5.5 are typically recovered by high-speed centrifugation at a laboratory scale, which appears to impede process scale-up. Our objective was to investigate the effect of flocculants on separation of precipitated proteins from process water (supernatant). Flocculants with different surface charge properties and molecular weights (Mw) were added to precipitated proteins. Protein separation was evaluated by determining optical density (OD) of the supernatant using Bradford dye-binding method. A high Mw anionic flocculent at 100 mg/L resulted in excellent protein separation following 10 min reaction. The OD of the supernatant was comparable to that of clear water, suggesting that even water-soluble fish muscle proteins were removed from the process water. Freeze-thaw cycles, commonly encountered in the fish processing industry, resulted in even more rapid flocculation reaction. This flocculent could be added to a bio-reactor that precipitates muscle proteins at pH 5.5 in a continuous isoelectric solubilization/precipitation system. However, effects of the flocculants on human and animal health should be determined and appropriate approvals obtained before the recovered muscle proteins can be used in human food products and/or animal feeds. 相似文献

This work is devoted to the biocides effects by electrochlorination under a pulsated bipolar polarization (PBP) of low potentials (− 1 V/cm to + 1.6 V/cm). Two working electrodes were used, one in platinum and the other in AISI 430 stainless steel, in two different electrolytes: sterilized distilled water and 0,9N sterilized NaCl solution, contaminated by E. Coli or by Pseudomonas Aeruginosa. The inactivation of these bacteria was established under the action of three biocide treatments: E.F (Electric Field); the combination of E.F and Active Chlorine (A.C) and the combination of E.F, A.C and Ions in Solutions (I.S). The synergistic effect of these treatments leads to a high efficiency in disinfection. The results showed that the total inactivation of the E. Coli stain is reached in half a minute, while that of P. aeruginosa is reached in 20 min. The use of an economic stainless steel electrode allowed us to observe that, for pulsate frequencies higher than 103 s− 1, the formation of active chlorine increased whereas the corrosion speed decreased. 相似文献

A novel MgO-type expansive agent in a designed formula has been prepared with industrial by-products—magnesite, serpentine and dolomite calcined at 1150 °C for 1 h. The compositions of expansive agent were studied by X-ray diffractometry (XRD), DSC and scanning electron microscopy (SEM), and the results showed that the main phases were MgO, CaO and C2S. The hydration of CaO to Ca(OH)2 is quick and can be completed in 3 days in water at 20 °C, and MgO to Mg(OH)2 is slow and about 57% MgO hydrates into Mg(OH)2 in 180 days under the same condition, which is quicker than in water. The hydrations of CaO and MgO generate expansions in early and later age, respectively. 相似文献

This paper summarizes studies on the presence of acid-fast and yeast organisms in wastewater and water treatment plants and in surface water. These organisms were found to satisfy three of Bonde's criteria for indicator organisms: presence whenever pathogens are likely to be present; resistance at least equal to that of pathogens; and lack of regrowth in the post-treatment environment.
